The new weekly train will start from Varanasi every Friday and from Vadodara every Wednesday.
The Mahamana Express route - a 1,531-kilometres journey between the two cities - will take 27 hours and 30 minutes.
The train offers ergonomically-designed ladders for climbing to upper berths and snack tables for side berths.
Its windows have powered venetian blinds and roller blinds. There are LED lights for reading and at several points to make it easier to find berths.
All coaches have fire extinguishers and the pantry car has an electrically-operated chimney.
The train will stop at Bharuch and Surat in Gujarat; Amalner and Bhusawal in Maharashtra; Itarsi, Jabalpur, Katni and Satna in Madhya Pradesh and Chheoki in Uttar Pradesh.
The Mahamana Express will have one AC First Class coach, two Second Class AC coaches, eight sleeper and four general coaches, one pantry car and two guard brake vans.
According to a statement, the train's toilets have platform washbasins, bigger mirrors and an odour control system.
The first Mahamana Express, named after freedom fighter Madan Mohan Malviya who was known as Mahamana Pandit, was launched last year.
The service currently runs in the Varanasi-Delhi and Bhopal-Khajuraho routes.
